1. Pellet 1–10ml of bacterial cell sample.
   (<5ml for high-copy plasmid, <10ml for low-copy plasmid). Discard supernatant.
   Add 200μl of Resuspension Buffer to the pellet and mix well by pipetting up- down until solution is homogenous.

2. Add 200μl of Lysis Buffer and mix well by pipetting up-down (5–10x) until solution is homogenous.
   Incubate for 8 mins at room temp.

3. Add 100μl of Neutralization Buffer and mix well by pipetting up-down (5–10x).
   Incubate for 2 mins at room temp.

4. Place tube on magnetic rack for 1 min.
   Transfer supernatant to a new microfuge tube. Discard previous tube.

5. Add 400μl of Binding Buffer to the supernatant in the new microfuge tube and mix well by pipetting up-down (10-15x) until solution is homogenous.
   Incubate for 1 min at room temp.

6. Place microfuge tube on magnetic rack for 1 min. Discard supernatant.

7. Add 600μl of Wash Buffer #1 and mix well by pipetting up-down (5–10x) until solution is homogenous.
   Place microfuge tube on magnetic rack for 1 min. Discard supernatant.

8. Repeat with 600μl of Wash Buffer #2, place on magnetic rack for 1 min.
   Discard supernatant and leave to dry for 1–5 mins.

9. Add 50–200μl of Elution Buffer and mix well by pipetting up-down (10–15x) until solution is homoegenous.
   Incubate for 1 min.

10. Place microfuge tube on magnetic rack for 1 min.

11. Transfer pure plasmid supernatant to a clean microfuge tube.
