Instructions for Use
TPP Tissue Culture Flasks
90151 Tissue Culture Flasks
TPP tissue culture flasks are intended for tissue culture in manual use. For
optimal cell adhesion and growth, only the bottom of the tissue culture flask
is opto-mechanically activated. The angled neck reduces the risk of medium
contacting the inside of the screw cap by spillage and allows excellent access
for cell scrapers and serological pipettes.
TPP tissue culture flasks are available with filter or VENT screw caps.
The tissue culture flask is for single use only.
Note:
Follow the national regulations for handling biological materials and wear the
appropriate protective clothing.
During the work process, be sure to observe the rules of aseptic technique.
Instructions:
- Inspect packaging and product for integrity.
- Use only materials in perfect condition.
- Check the expiry date (EXP) on the label and packaging. Use only products with a valid EXP.
- Open the package in a sterile environment and remove a complete system for use Open the flask and fill it with the medium and inoculum according to your laboratory routine. Please refer to the optimal filling volume (see technical data).
Filter Screw Cap:
- Close the filter screw cap with ventilation holes. Continuous gas exchange occurs through the integrated 0.22 µm hydrophobic membrane (Figure 1). If the membrane becomes wet, gas exchange is temporarily reduced.
VENT Screw Cap:

For a gas exchange, turn the VENT cap into the ventilation position at 12 o’clock. You will hear/notice a click.

Visual check: Rectangle at 12 o’clock (Position 1)
During incubation of cells, the rectangle must be in position 1 / 12 o’clock. -
To stop gas exchange, rotate the cap 1/4 turn clockwise beyond the vent position. The flask is now tightly sealed and there is no gas exchange.
• Visual check:
Rectangle at 3 o’clock. (Position 2) -
For a transfer of the flask within in the laboratory (laminar flow ↔incubator), TPP recommends closing the VENT screw cap tightly.

Cultivate and harvest cells according to your common laboratory routine.

In order to obtain uniform cell growth, the following points must be observed:
o Resuspend the cells well in the medium to avoid cell clumping.
o Avoid foaming during resuspension and seeding to avoid holes in the cell layer.
o Low volume of medium leads to meniscus formation and thus to uneven cell growth (cell clumps at the edge).
Please refer to the technical data for the optimal filling volume. -
Air vents in the bottom rim ensure optimum heat distribution in the incubator when several flasks are stacked on top of each other.

Test the many influencing factors in advance under routine conditions.

Store at room temperature before use, protected from UV light.
Advice and Recommendations:
- For labelling there are bilateral inscription fields
- The volume scale is an indication of the filling volume, not an absolute value.
- The stacking rim allows a stable stacking of several flasks on top of each other, also in combination with other TPP flasks of the same size.
- For homogeneous cultivation of cells, it is essential to level the incubator and provide a continuous supply of medium.
- Avoid and/or eliminate sources of vibration near the incubator to ensure uniform cell growth.
