TPP 90028 Tissue Culture Flasks with Peel-off Foil
Instructions for Use
TPP tissue culture flasks with peel-off foil are designed for cell culture with necessary sterile handling techniques, storage and microscopy in manual use and to meet the need for a large one-time access from the above to the cell culture. The growth surface is optomechanically activated to optimize cell adhesion and proliferation. The angled flask neck reduces the risk of medium contact with the inside of the screw cap. TPP cell culture flasks with peel-off foil are available with filter screw caps only, when using a VENT screw cap a possible pressure build-up of 0.03 bar or higher could press open the peel-off foil. The TPP tissue culture flask with peel-off foil is intended for single use only.
Note: Follow national regulations when handling biological material, use appropriate protective clothing Take note of the regulations of aseptic working during the work process.
Instructions:
- Check the packaging material and the product for intactness. Use only faultless material.
- Check the expiry date (EXP) on the label and packaging. Only use products with valid EX
- Open the packaging in a sterile environment and remove a complete system for use.
- Open the filter screw cap of the cell culture flask and fill the flask with medium and inoculum according to your standard laboratory routine.
- Close the filter screw cap completely. A continuous gas exchange takes place through the integrated hydrophobic membrane0.22 µm. If the PTFE filter membrane gets wet, the gas exchange is reduced for a short time.
- Cultivate and harvest cells according to you common laboratory routine.
- To achieve an uniform cell growth, the following aspects must be observed:
- Resuspend the cells well with the medium to avoid cell lumps.
- Avoid foaming during resuspension and seeding to avoid holes in the cell layer.
- Low medium volume leads to meniscus formation and thus to uneven growth of the cells (cell clumps at the edge). Please refer to the technical data for the optimum filling volume.
- Air vents in the bottom rim ensure optimum heat distribution in the incubator when several flasks are stacked on top of each other.
- The mechanical strength of the flasks during centrifugation is influenced by
- Shape and material
- Accuracy of fit in the centrifuge adapter
- Temperature, centrifugation time, g-number
- Chemical and physical properties of the centrifuged material
- Rotor type: fixed-angle or swing-out roto
- Test the multitude of influencing factors under routine conditions beforehand.
- Storage before use at room temperature, protect from UV light.
Peel-off the Peel-Off Foil
- The cell harvest from chamber can be carried out from above when the peel-off foil is removed.
- Position thumb and middle finger on the inscription fields on both sides of the neck of the T-flask. Use your index finger to press the flask onto the surface by placing the fingertip underneath the foil flab, thus stabilizing the flask. Grab the foil flap with the other hand and pull the foil back with a gentle pull.
- It is not possible to reclose the foil.
Advice and Recommendations
- For labelling there are inscription fields on both sides of the flask.
- The graduation serves as a reference for the filling quantity and is not an absolute value.
- The stacking rim allows a stable stacking of several flasks on top of each other, also in combination with other TPP flasks of the same size.
- For a homogenous cultivation of the cells, levelling of the incubator and continuous supply of medium is essential.
- Avoid and/or eliminate vibration sources near the incubator to ensure uniform cell growth
Note: The barrier limits the growth area to 115 cm2.
