Instruction for Use
TPP Tissue Culture Test Plate
TPP tissue culture plates are designed for the cultivation and growth of cells
as well as for photometric measurements and cell-based assays. The growth
areas – but not the sidewall of the wells – are optomechanical activated for
optimal cell adhesion and growth.
The tissue culture test plate is intended for single use only.
Note:
Follow national regulations when handling biological material, use appropriate
protective clothing
Take note of the regulations of aseptic working during the work process.
Instructions:
- Check the packaging material and the product for intactness. Use only faultless material.
- Check the expiry date (EXP) on label and packaging. Only use products with valid EXP.
- Open the packaging in a sterile environment and remove a complete system (lid with plate) for use.
- Open the plate and fill it with the medium and inoculum according to the usual laboratory routine.
- Cover the wells with the supplied lid to reduce the risk of cross-contamination.
- Cultivate and harvest the cells according to the usual laboratory routine.
- In order to achieve uniform growth on the surface, the following aspects must be considered:
- Resuspend the cells to avoid cell clumps.(Uneven growth)
- Avoid foaming during resuspension creating holes in the cell lawn.
- Low medium volume leads to the formation of minuscule cells, which leads to uneven growth of the cells.(many cells on the edge, few in the middle).
- For optimum filling, volume refer to the technical data.
- The air vents in the bottom rim allow even heat distribution in the incubator even when several plates are stacked on top of each other. Spacer cams on the inside of the lid ensure constant gas and moisture exchange with minimal evaporation. This eliminates the need to fill the space between the wells.
- Identification: the black alphanumeric inscription of the wells, yellow inscription field on lid and plate.
- Centrifuging: The use of suitable rotors or centrifuge adapters is recommended. Follow the safety instructions of the centrifuge manufacturer. See also TechDoc.
- The mechanical strength of the plates during centrifugation is influenced by
- Shape and material
- Accuracy of fit in the centrifuge adapter
- Temperature, centrifugation time, g- number
- Chemical and physical properties of the centrifuged material
- Rotor type: fixed-angle or swing-out rotor
- Test the multitude of influencing factors under routine conditions beforehand.
- Storage before use at room temperature, protect from UV light.
Advice and Recommendations
- The stacking rim enables safe stacking of several plates, also in combination with other TPP plates of different well number.
- For homogenous cultivation of the cells, leveling of the incubator and continuous supply of medium is essential.
- Avoid and/or eliminate vibration sources near the incubator to ensure uniform cell growth.
ELISA : The TPP plates feature is not suited for use in ELISA tests.
