Product Usage Instructions
Program Setup:
- Program your real-time PCR instrument with the following channels: FAM (peanut) and HEX (Internal Control).
- Set the cycling parameters as follows:
- 4 min at 95°C
- 10 min at 60°C
- 50 cycles of 5 sec at 95°C and 60 sec at 60°C with fluorescence detection
Data Interpretation:
- Verify results of positive (Control Template) and negative controls (H2O) before interpreting sample results.
- Always compare sample results to the positive and negative controls.
- Interpret results based on the table provided in the manual.
Preparation of the PCR Mix:
- Take precautions to prevent contamination by using filter tips and gloves.
- Thaw reagents, mix without vortexing, and spin vials briefly before opening.
- Add PCR mix to wells.
- Add samples and controls.
- Optionally, prepare a standard curve for quantification referring to the Allergen RM 800 product manual.
- Seal strips/plates accurately.
- Briefly spin strips/plate in a suitable centrifuge.
- Start the real-time PCR run following the cycling parameters set up earlier.

PROGRAM SETUP
Program your real-time PCR instrument before setting up the PCR reactions. Select the following channels:
- FAM (peanut) and HEX (Internal Control).
For some real-time PCR instruments, the probe quencher as well as the use of a passive reference dye must be specified. This kit contains probes with a non- fluorescent “dark” quencher and no passive reference dye.
DATA INTERPRETATION
Verify results of positive (Control Template) and negative controls (H2O), before interpreting sample results. Always compare samples to positive and negative control. Review data from each channel and interpret results as described in the table.

PREPARATION OF THE PCR MIX
Take appropriate precautions to prevent contamination, e.g., by using filter tips and wearing gloves. Thaw reagents, mix (do not vortex!) and briefly spin vials before opening.

ADD PCR MIX Pipet 20 μL of Master Mix into each strip or plate well (n samples + 2 controls).

ADD SAMPLES AND CONTROLS Pipet 5 μL of samples, Negative Control (colorless cap) or Control Template (purple cap) into respective wells.
OPTIONAL:
STANDARD CURVEFor quantifi cation, prepare the standard curve. Please refer to the Allergen RM 800 (Product No. KIT230009) product manual. -
