  1. Check the voltage specified on the name plate and ensure it matches the line voltage in your location.
  2. Please c he c k t h e sample solution and the material of t he instrument with c h emical compatibility chart before using.
  3. Do not place the instrument nearby any flammable o r explosive materials. Install the instrument in a clean, dust less and ventilated area.
  4. During the operation, overpressure or t hick liquid w ill cause larger loading to the moto r module, the motor module will stop automatically for s elf protection. Please turn off the power and turn it on again after 5 minutes.
  5. Please clean the tubes and appearance of t he instrument after using it.
  6. If you have any problem s with using the instrument, pleas e contact the service crew for help . DO NOT disassemble improperly.
  7. Please discard packaging materials in accordance with relevant local regulations

Pressure in tubing should be less than 40 psi, or the tubing would be expanded due to over pressure.
Please turn off and unplug the instrument before changing the fuse.

Please check if the package is complete without an y damage before unpacking. When unpacking, please make sure you have all a cc e ssories that indicated on the list. Installation

Support Base Set

  • To install the support base set on the platform of case u sing a hex wrench to fasten and fix the 2 hex socket head cap screws.
  • Please notice the direction of sup port rod , the machining plane of support rod is facing to the aft side.

Gauge Mounting Set

  • Install the feed gauge mounting set thro ugh the support rod with gauge facing forward fasten an dtighten the set with hand screw on the back
  • In stall the cap sulebracket into the base for capsule bracket . Then install this assemble through the supportrod , fasten and fix it on to t he middle of the support rod with hand screw
  • Install the ret entategauge mounting set thro ugh the support rod with gauge facing forward fasten and fix the set on to the upper side of support rod with hand screw.

Magnetic Stirrer and Reservoi

  • Place the magnetic stirrer in the groove of the plat form and plug the AC socket on the backw ard of Tanfil 100 Install the filter discon to the lit , and put the reservoir (with lid) on to the magn etic stirrer.

Three Way Stopcock

  • Install the three ways topcocks to the retenta te gauge mounting blockfeed gauge mo unting block and res ervoir . Refer to 3. Main Part Diagram for the installation positions.

Tubing

  • Please cut the tubing and install the luer connect ors a ccording to the foll owing information.

  • Specification of luer connectors dep end s on the capsule.
  • After install ing the tubing, please install the screw clamp to the concentrate tubing.

Tubing Position

Operation

  • There are several op eratio n procedures in this syste m , please follow the steps below tos etup the flow path you need For cleaning pro cedure and solution for capsules , please refer to the instruction manuals of cap sules.

Cleaning

  • Please clean the system with the following steps before first use Use the cleaning tub ingtoconnect the feed and retentate gauge moun t ing blo cks , asshown in figure 1.
  • Pour the cleaning solution in to the reservoir and capit with the lid kit
  • Set the three way stopco cks and adjust the flow path as shown in figure 1.
  • Turn on the pump, increase the spee dslowly to 50 rpm.
  • Please check if there is any le akage . Make sure to fasten all the connectors.
  • Let the sys tem circulate for 15~30 minutes, and turn off the pump.

Draining and Collection

  • When the pump is stopped setting the three way stop coks as shown in figure 2

  • Open one of the lid cap son the reser voirl id the solution  ould flow through the recovery tubing to the container automatically by gravity.

  • If you need to collect the solution in the system after experiment, please turn off the pump first and adjust the three way stopco cks, then exchange the capsule to the cle aning tub ing as shown in figure 3.

  • Turn on the pump to collect all the solution in to the reservoir and turn off the pump after collect ion The f low path refers to figure 4.

  • Setting the three way stop cock at the bottom of the reservoir to adjust the flow pat has shown in figure 5 Open one of the lid capson the reservoir lid the solution would drain automatically by gravity.

  • To drain the solution compl etely , please set the three way stopco ck of the r eservoi r and a djust the flow path as shown in figure 6 . Turn on the pump to drain the soluti on.

  • Tubing can be removed for high temperature sterilization as require d after first use and cleaning.

Capsule Rinsing

Rinsing Capsule

  • Set the three way stopcocks and adjust the flow path as shown in figur e 7 Pour the rinsing solution in to the reservoir.
  • Turn on the pump Increa se the speed (rpm) slowly un til the feed pressureac hieves to the suggesedvalue that depends on the capsule
  • Please check if there is any leakage . Make sure to f asten all the connector s. c. C i rculate the rinsing so lu tion continuo usl y T he ri nsing so lution vo lum e refer s to the op eration manuals of t he capsule.

Rinsing the Capsule and the System

  • Set the three way stopcock s and adjust t he flow path as shown in figure 8
  • Turn on the pump . Inc re ase the speed (rp m) slowly until the feed pressure achieve s to the suggest edvlue that depend son the capsule
  • Please check if there is any leakage . Make sure to fasten all the connectors.
  • Adjust the screw clamp and the speed ( rpm) of pump unti l the retentate and feed pressure achieve the suggested value that depends on the capsule
  • Circulate the rinsing solution continuouslyu ntil the volume of the solu tion in reserv oir le ft a s sug ges ted. Turn off the pump, and refer to 2 Draining a nd Colle cting ste psas mentio ned before to drain the solution.
  • Please clean the caps ule bef o re resin ingit . The cleani ng method of caps ule depen dson its own operation manual.

Pretreatment and Concentration

Pretreatment

  • Set the th ree way stopcock s and adjust the flow path as shown in figure 9
  • Pour the buffer in to the rese rvo ir Turn on the pump a nd inc rease the speed (rpm) slowly to fill the tube with out bubble s T he feed pressure depends on capsulei tself
  • Please check if the re is any leakage M ake sure to fasten a ll the connectors.
  • Stop the pump just as th evolume in the reser voir reaches the bott om.
  • Dis assemble the filtrate tubing to pour out all the liquid in the tube then assemble the tubing back to the caps ule to complet e the pretreatment.

Concentration

  • Pour the sample s olution into the reservoir , turn on the pump. T he feed press ure depends on capsuleitself
  • Adjust the screw clamp and the speed (rpm) of pump until the fe ed pressu re and retent ate pressure ach ieve to the pressure as sug gested Sugges ted pressure depends on caps ule itself.
  • The feed pressure should NOT be greater than the sugg ested pressure of the capsule and the retent at epressure shou ld NOT be g r eater tha n 40 psi
  • Cont inu ous circul at ion for the e xpected concentrat ion of the solution. A nd turn o ff the pump.
  • Refer to “ 2 Draining and Collecting ” step s a s mentioned before to collect all the s amp l e solution in to the container

Continuous Sampling

  • Samples over 500 mL could be put in other flasks. Use a silicone tube to connect the reservoir lid and the flask as shown in figure 10 . During t he diafiltration , the system will b e come negative pressure gradually. Thus, the solution in the flask will transfer to the reservoir automatically.

Usage and Maintenance of Capsule

  • In this manual, we take Pall Minimate TFF Capsuleas example. For other capsules, please refer to manufacture s instruc tion manuals.

Using the Capsule for the First Time

Flushing

  • Pour about 500 mL deioni z ed wat er into the reservoir. Slowly adjust the flow rate until the feed pressure reaches 2 bar (a bout 30 p si)
  • Place the tubing from the reten tate and filtrate ports into a waste container.
  • Slowly increa se the retent a te press ure by screw clampu ntil the retent a te flow rate i s approxi mately eq ual to the filtr at eflow rate. Both tubing should drain about 250 mL wate r individually.
  • Filtrate flow rate is relatively slow whenusing a smaller Molecular Weight Cut Off MWCO capsule . Flus hing a bou t 500 m L wate ris enough in princip le.

Determining the Original Normalized Water Permeability NWP of Capsule

  • After flushing, pour deionized water into the reservoir.
  • Close retent ate port and slowly increase the speed (rpm) unt il the feed pressure reach es 1 bar ( a bout 15 p si
  • Record the volume of drain from filtrate port in 10 minutes. And converts the data to filtrate flow rate (mL/min).
  • You can measure the volume of drain by (1)bal ance for weight or 2) graduated cylinder for volume.

Cleaning after Using

  • Choosing the Cleaning Solution
  • Using 30~45°C s olu tion to enhance the c leaning effec t
  • 0.1 N~ 0. 5 N N aOH
  • 0.1 N Nitric acid
  • 0.1 N Ace tic acid
  • 0. 1 N Phosphoric acid

Cleaning Steps

  • Drain the Waste: A fter recover the sam ple , place the tubing from the reten tate and filtrate ports into a waste container.
  • Flushing with Deio nized Water : Refer to (1) I Flushing.
  • Cleaning with Cleaning Solution : follow the steps below to clean the capsule .
  • Make sure that the feed pressure should NOT ex ceed 2 bar (about 3 0 psi).
  • Place abou t 500 mL cleaning solution into the reservoir. Open the ret entateport and close the filt rate portreten ta te for abo ut 30 minutes.
  • Change 5 00 mL of new cleaning solution , retenta t e 30 minutes and drain the solution.
  • Change 500 m L of new cleaning solution Close retentate port and open filtrate port, pump about 250 mL 3 00 mL of solutionthrough filt rate tubing.
  • Determ ining the Membrane Recovery : Repeat 1 ) II NWP test to evalu ate the condition of capsule.

Membrane Recovery = NWP (after cleaning) / NWP original)

  • Membrane recovery < 75 % Clean the caps ule again.
  • Membrane recovery < 5 0% Replace with a new capsule

Storage of Capsule

Before storage, please make sure you clean the caps ule as 2) II Cl eaning Ste ps every time.

< 6 months 0.05N ~ 0.1N sodium hydroxide (NaOH)
> 6 months 15 % glycerin + 0.05 % sodium azide (NaN3)
  • Flush ing the capsule before using every time. ( Refer to (1) I . Flushing.

Maintenance

  1. Please unplug the instrument before cleaning it.
  2. Please operate the instrument in well ventilated area and keep it clean all the time. If there is any solvent splash to the case, please wipe it off to avoid the damage caused by corrosion.
  3. O-rings are consumable materials that should be replaced annually. If there is any functional disorder, please replace it as soon as possible.
  4. Silicone tubes are consumable materials. If the tubes broke or expand during the operation, please replace it as soon as possible.
  5. If the fuses burnt, please troubleshoot first. And use flathead screwdriver to screw up the fuse holder. The fuse holder is equipped with spare fuse.

Troubleshooting

Issue Cause and Solution

Abnormal Display

| Loose plug → Reconnect plug to power supply. Fuse burn → Replace a new fuse.

Display or components failure → Contact distributor for assistance.

Suddenly Stop of

Motor during Operation

| ****

Overload of pump module → Slow down the speed and loosen the screw clamp. Restart the instrument.

No Stir with Magnetic Stirrer| Loose plug → Reconnect plug to power supply.

Disorder of magnetic stirrer → Contact distributor for assistance.

Expand of Silicone Tube during Operation| Overpressure of silicone tube → Slow down the speed and loosen the screw clamp. Replace with a new silicone tube.

Capsule block → Replace with a new capsule.

Others| Contact distributor for assistance.
