ALPCO Multi Array Xplorer 45k MAX 45k Fully Automated Multiplex Batch Analyzer Instructions

• The MAX 45k enables sample-to-answer testing for 295 allergen-specific IgE antibodies (ALEX2) or 286 food-specific IgG antibodies (FOX). Testing for both panels can be completed with just 500 µl of blood from a finger prick or blood draw for a complete profile.
• The ALEX2 allergy panel incorporates 183 molecular allergens – most in the industry. Well-characterized single molecular components provide more specific information regarding potential allergic response (specific allergen vs. allergen family) and improve batch-to-batch consistency.

Instrument Features

• The cartridge rotor ❶ holds up to 50 cartridges for multiplex analysis of up to 50 samples in a single 4-hour run.
• The wash dispensing needle ❷ adds wash buffer to individual cartridges as the rotor turns.
• The Imaging Module ❸ scans each cartridge to verify cartridge type, lot number, expiration date and position and captures an image of each cartridge at the end of the assay.
• The steel needle pipetting arm ❹ moves up and down and pivots to transfer from the sample and reagent rotor to the cartridge rotor.
• The active wash station ❺ thoroughly washes the pipetting tip between each sample and reagent transfer preventing cross-contamination and eliminating the need for disposable tips.
• The Sample Rotor can accommodate up to 50 samples ❻ and reagents for a 50 sample ALEX2 or FOX run. ❼ Reagents are inserted with bar codes facing out and read by the MAX 45k. ❽

Typical Workflow

Step 1: Load Reagents

Load assay-specific reagents with bar codes facing out. Ensure wash buffer reservoir is filled and waste reservoir is empty.

Step 2: Load Samples

Load samples with bar codes facing out. 400 µL is required for standard sample tubes for ALEX2. 200 µL is sufficient for false-bottom, low volume tubes. Samples are pre-diluted using 12 µL of sample for FOX testing. Vial sizes can be mixed.

Step 3: Load Cartridges

Cartridges are loaded from the center pushing outward until a clicking sound is heard beginning at Position 1.

Step 4: Close Lids and Manage Inventory

During the inventory management, the MAX 45k will record reagent, sample, and cartridge information from bar codes and log positions.

Step 5: Make Priming and Predilution Selections and Verify Reagent Volumes

Select normal or extended priming and indicate if all samples have been prediluted. During verification, the MAX 45k will confirm sufficient volumes for all assay reagents.

Step 6: Start the Run

The MAX 45k will automatically perform all dilution, pipetting, incubation, wash, and cartridge imaging steps. When the assay is finished, the status field will indicate “Test Run Finished”. By clicking on “View Measurements”, the result page is opened.

Array Panels

ALEX2 Allergy Xplorer:
~300 allergens, including 183 molecular allergens

• The ALEX2 panel incorporates 144 recombinant molecular allergens and a powerful blocking agent to prevent nonspecific binding and potential false positives due to reactive sugars (CCDs or cross-reactive carbohydrate determinants) on some allergens for more accurate results.

FOX Food Xplorer:
283 foods covering 13 food groups

• The FOX panel measures all specific IgG antibodies, including immune-activating IgG1 and IgG3 subclasses, immune-modulating IgG2 antibodies, and immune-blocking IgG4 antibodies for detection of potential food intolerance or food tolerance.
