Intended purpose
Preparing, storing and transferring human embryos. To be used only with the MIRI® TL device.
About the product
The Esco Medical CultureCoin® is a sterile, single-use culture vessel
specifically developed for use in IVF. It consists of two parts: dish and
cover. The dish holds up to 14 embryos in the centrally placed line of culture
wells. The culture well features a 300µm, an optically clear area where the
embryo is situated. The area is optimized for microscopy. A clear circle is
present around the culture area for the purpose of making it easy for an
automatic focus algorithm to find the area under microscopic image
acquisition. The culture wells are numbered (1 – 14) for easy location of each
well by the user. The dish has 4 washing wells for cell manipulations, washing
media or replacement media.
A large well is provided outside the culture area for pH measuring without
affecting the cultured samples. The dish has a flat bottom, so it is ideal for
heat transfer from below.
The cover is mounted on top of the dish, covering the whole top area
completely. The user will only grab the dish during handling by pinching on
the grip-lip features of the dish without the risk of losing the dish by
holding on to the cover only – as can be the case on all traditionally
designed dishes. Mode of action: Esco Medical CultureCoin® was designed to
ensure a safe and intended environment to help treat patient infertility
problems and assist with the conception of a child. Embryo growth in the
CultureCoin®
is achieved using specific non-toxically fertility or reproductive function
plastic, media that provide a nurturing environment, and the MIRI® TL family’s
multiroom IVF incubator that creates and keeps stable environment temperature
and gases concentration.
Principles of operation of the device: CultureCoin® is designed to culture
cells only in Esco Medical Technologies MIRI® TL family’s multiroom IVF
incubators for a period of up to 6 days. The capacity of droplets in
incubation wells is 25μl. Such an amount of media is sufficient for cell
development during such a period. A high wall surrounds the droplet area to
ensure enough space for an oil overlay to secure media from evaporation.
Incubation wells are situated in one line, at the same height and across the
exact distance between well centers. It perfectly fits the design of the MIRI®
TL family’s multiroom IVF incubators, which camera moves under CultureCoin®
along incubation wells.
The CultureCoin® dishes are packed individually. The packing box contains 25
dishes.
Model Code: MRI-CC. Ref No: 1821493.
Only individuals with formal education in a relevant field of healthcare or
medical discipline could work with CultureCoin® dishes.
The CultureCoin® ‘s intended target groups are IVF patients. Patients are
women in their reproductive years who have fertility health issues. The
intended target group indication is IVF treatment. There are no intended
target group contraindications. CultureCoin® is a medical device intended to
be used on one patient during a single procedure. It is not intended to be
reprocessed (cleaned, disinfected/sterilized) and used on another patient.
CultureCoin® can be used only with the MIRI® TL family’s multiroom IVF
incubators and sequential media specifically designed for IVF treatment.
CultureCoin® cannot be used with non-medical devices and other non-IVF
treatment-designed media.
The device is manufactured under a full EU-certified 13485 ISO quality
management system.
This product conforms to the requirements of the Regulation (EU) 2017/745
concerningmedical devices and is classified as a Class IIa device under rule
II.
Personal Protective Equipment (89/686/EEC) and Machine Directive (2006/42/EC)

6.1 Pre-use
- Do not use the product if the package is damaged. Contact Esco Medical Technologies or the local Representative.
- Always keep these instructions in a place where they can be easily accessed.
With CultureCoin® dishes could work only individuals with formal education in
a relevant field of healthcare or medical discipline.
Basic training on how to use CultureCoin® with MIRI® TL family’s multiroom IVF
incubators is provided along with the installation of MIRI® TL family’s
multiroom IVF incubators.
6.2 During use
- Use only for the purpose listed under the intended purpose.
- The dishes are single-use only. Please do not attempt to clean and reuse them.
- Only for the use of water-based culture media, mineral or Paraffin oil.
- Do not use this product in the MIRI® TL family’s multiroom IVF incubators at temperatures exceeding 40.0 °C.
Functions related to SAFETY:
- Identifying the correct dish;
- Picking and lifting the dish;
- Avoiding cross-contamination;
- Placing the embryo correctly.
Please refer to sections 9 – 14 in this manual for more information on
properly using CultureCoin®.
6.3 Post-use
If the dish was used to store/incubate biological samples, it should be
discarded as biohazardous waste according to laws and procedures valid in the
user’s country.
The Culture mode
The CultureCoin® can be used only for culturing with culture media filled into the wells and oil overlay. They cannot be used for open culture.
Overview
The CultureCoin® dish consists of 2 parts:
- CultureCoin® dish (1).
- Cover (2) that covers the entire CultureCoin® area.
Use
- Unpack both the cover and dish in an aseptic environment.
- Leave the dishes with the covers on for off-gassing in the aseptic environment for 12 hours (overnight).
- Warm the dishes on a heated stage or an incubator before filling them with media.
- Fill all the culture wells with approximately 25 µl of precultured equilibrated culture media in an aseptic environment.
- Filling of the washing wells is optional. One washing well contain approximately 23 µl. They can be used for cell manipulations, washing media or replacement media.
- Check under a microscope for any visible air bubbles and remove them with a stripper tip.
- Fill the oil area with a confluent layer of suitable mineral or Paraffin oil to cover the culture media. The recommended amount is 5 – 5.5 ml. Do not overfill.
- Put the cover on the dish and leave it for 4 hours of equilibration in the CO2 incubator (humidified incubator is preferable).
- Check for air bubbles in the culture wells after the equilibration. Remove any bubbles with a stripper tip.
- Load the embryos in the culture wells. Make sure the embryos are located very precisely in the center of the culturing well and not on the side slope.
- Optionally fill the pH well with culture media up to the lowest step (recommended volume – 350 µl) of the pH well and cover it with an oil overlay (recommended volume – 150 µl).
- Put on the CultureCoin® cover.
- Put the CultureCoin® in the MIRI® TL family’s multiroom IVF incubator as instructed in the 10 th section below.
CultureCoin® placement in the compartment
Before starting the time-lapse, correctly placing the CultureCoin® in the compartment is essential. To ensure the correct position of the CultureCoin® (so that the camera can identify all the culture wells), place the CultureCoin® in its place and secure its position by pushing it downwards and towards you, then to the left edge.
Removing the CultureCoin® cover
CultureCoin® cover does not have a lot of free movement on the dish while it
is closed, but the cover may move slightly during transportation. The cover
can rotate a bit on the CultureCoin® and get stuck on the dish edges. As a
result, it may be harder to open it.
There is a specific technique for the CultureCoin® cover opening in such a
case. Hold the dish on the grip-lip and lift the cover from the cut
edge.
The use illustrated
Each of the 14 culture wells is filled with approximately 25 µl culture
medium. The embryo is placed at the bottom of the culturing area, as shown in
Figure 12.2.
The washing wells can also be filled with 23 µl of culture media, but it is
not a requirement. The culture media is filled into the culture
wells (marked red), then oil (marked bluegreen) is added to cover the entire
cultivation area.
pH measuring
Validating the pH of the culture media should be a standard procedure. The
CultureCoin® has a large well outside the culturing area (Figure 8.1). It
makes it possible to make spot checks safely and conveniently on the pH level
without disturbing culture wells.
The pH is measured using this procedure:
- Calibrate the pH electrode following the “User manual of MIRI® TL family’s multiroom IVF incubators”, section 18.
- Remove the oil overlay from the pH well.
- Insert the pH electrode, inspect that the electrode sensor is fully covered with the culture media.
- Measure pH in the culture media.
- If pH will be measured after some time again, do not forget to fill an oil overlay under the culture media (recommended volume – 150 µl).
- Repeat the pH measurement procedure from Point 1.
Adjust the CO2 level if necessary.
When choosing a pH electrode, it is necessary to consider the probe’s size.
Esco Medical Technologies recommends using a 3 mm diameter probe size.
Writing area on the CultureCoin® and lid
There is a dedicated area where the user can write or put a label with patient
data on the CultureCoin® dish area marked red in Figure 14.1 below.
Only use special markers that do not release volatile organic compounds
(VOCs).
