Safety Instructions

General safety instructions

Please read carefully!

The instrument seripettor® can be used in combination with hazardous materials, work processes and equipment. However, the operating manual cannot cover all of the safety issues that may occur in doing so. It is the user’s responsibility to ensure compliance with the safety and health regulations and to specify the corresponding restrictions before use.

  1. Every user must read and understand this operating manual before operation.
  2. Follow the general hazard instructions and safety regulations (e.g., wear protective clothing, eye protection, and protective gloves).
  3. Observe all specifications provided by reagent manufacturers.
  4. When dispensing inflammable media, make sure to avoid to buildup of static charge, e.g., do not dispense into plastic vessels and do not wipe instruments with a dry cloth.
  5. Use the instrument only for dispensing liquids, with strict regard to the defined limitations of use and operating limitations. Comply with the operating exclusions(see Operating Exclusions,)! If in doubt, contact the manufacturer or supplier.
  6. Always perform work in a manner that does not endanger yourself or other people. When dispensing, the discharge tube must always point away from you or any other person. Avoid splattering. Only use suitable vessels.
  7. Never press down the piston when the discharge tube is sealed with the closure cap/screw cap.
  8. During operation, the piston locking mechanism must be closed.
  9. Never remove the discharge tube while the dispensing cylinder is filled.
  10. Reagents can accumulate in the closure cap/screw cap of the discharge tube. Therefore, clean regularly.
  11. For small bottles, and when using the flexible discharge tube, use a bottle stand to prevent tipping over.
  12. Never carry the mounted instrument by the actuating unit or the valve block, see First Steps, p. 47. Breakage or loosening of instrument parts can lead to personal injury from chemicals.
  13. Never use force. Always press the piston down gently when dispensing.
  14. Use only original accessories and original replacement parts. Do not make any technical modifications. Do not dismantle the instrument any further than is described in the operating manual!
  15. Always check that the instrument is in proper working condition before use. The user can come into contact with media if the instrument has been insufficiently cleaned or inspected. If there is a sign of a potential malfunction (e.g., piston difficult to move, sticking valves or leak age), immediately stop dispensing and consult Troubleshooting. Contact the manufacturer, if necessary.

Function

The seripettor® bottle-top dispenser is used for dispensing liquids directly from the storage bottle and is available in two versions:

Operation

When the instrument is correctly used, the dispensed liquid comes into contact with only the following chemically resistant materials:

  • PP, PE, borosilicate glass, Al2-O3-ceramic, ETFE, PFA, FEP, PTFE, Pt-Ir

Limitations of Use

This instrument is designed for dispensing liquids, observing the following physical limits:

  • Use between +15°C and +40°C of instrument and reagent (seripettor®: agar culture medium up to max. 60 °C)
  • Vapor pressure up to 500 mbar
  • Density up to 2.2 g/cm3
  • Kinematic viscosity:
    • 2 ml instrument: 300 mm2/s
    • 10 ml instrument: 150 mm2/s
    • 25 ml instrument: 75 mm2/s
  • dynamic viscosity [mPas] = kinematic viscosity [mm2/s] x density [g/cm3]

Operating Limitations

  • Liquids, which form deposits may make the piston difficult to move or may cause jamming (e.g., crystallizing solutions or concentrated alkaline solutions). If the piston movement becomes sluggish or stiff, the instrument should be cleaned immediately (Cleaning, p. 57).
  • When dispensing inflammable media, make sure to avoid to buildup of static charge, e.g., do not dispense into plastic vessels; do not wipe instruments with a dry cloth.
  • The instrument is designed for general laboratory applications. Compatibility of the instrument for a specific application (e.g., trace material analysis, food sector etc.) must be checked by the user. Approvals for specific applications, e.g. for production and administration of food, pharmaceuticals or cosmetics are not available.
Operating Exclusions

seripettor®

Never use seripettor® for:

  • Liquids attacking FEP, PP, PE or EPDM
  • Non-polar solvents such as hydrocarbons and halogen hydrocarbons
  • Concentrated or oxidizing acids
  • Explosive liquids
  • Carbon disulfide

seripettor® pro

Never use seripettor® pro for:

  • Liquids attacking PP, PE, Al2-O3-ceramic, ETFE, FEP, PFA and PTFE (e.g., dissolved sodium azide*)
  • Liquids attacking borosilicate glass (e.g., hydrofluoric acid)
  • Liquids which are decomposed catalytically by platinum-iridium (e.g., H2O2)
  • Non-polar solvents such as hydrocarbons and halogen hydrocarbons
  • Concentrated or oxidizing acids (except for HCl)
  • Explosive liquids
  • Carbon disulfide
  • Suspensions (e.g., activated carbon solutions) as solid particles may clog or damage the instrument
  • Dissolved sodium azide permitted up to a concentration of max. 0.1 %.

Storage Conditions

  • Store the unit and accessories in a cool and dry place in cleaned condition only.
  • Storage temperature from −20 to 50°C (−4 to 122°F).

Aqueous solutions

Biological buffer solutions and detergents, anti-foaming agents, culture media, vitamin solutions, etc. as well as hydrogen peroxide can be dispensed in everyday routines. Agar culture media can be dispensed up to max. 60 °C.

Acids

  • Low-concentrated or diluted, non-oxidizing acids can be filled.

Alkalis

  • Can be used for dispensing alkaline media such as NaOH, KOH and ammonia.

Polar solvents

  • E.g., ethanol, methanol, acetylacetone, etc.

seripettor® pro application range

The seripettor® pro-bottle-top dispenser expands the range of applications. It enables dispensing of acids (e.g., concentrated HCl), polar solvents (e.g., acetone) and UV-sensitive media.

Assembly

First Steps

WARNING: Follow during every use, especially with hazardous media

  • Wear protective clothing, eye protection and appropriate hand protection!
  • Never press down the piston when the screw cap is screwed on!
  • Avoid splashing the reagent!
  • To avoid splashes dispense slowly.
  • Liquid may accumulate in the screw cap. Open the screw cap slowly in order to prevent splashing.
  • Follow all safety instructions and comply with the operating exclusions and limitations of use; see Limitations of use and Operating exclusions.
  1. Screwing on the dispensing cartridge
    • a. Screw dispensing cartridge into valve block.
  2. Installing the actuating unit
    • a. Open piston lock on the actuating unit.
    • b. Put actuating unit over the dispensing cartridge and screw it down to the valve block.
    • c. Depress actuating unit up to the lower stop and keep holding it using one hand and with the other hand, close piston locking on the actuating unit.
    • d. Release actuating unit and check to see whether the piston in the dispensing cartridge has moved upward.

NOTICE: Check that the discharge tube is securely seated. Re-tighten cap nut(s) after two days of use.

Installing the filling tube

  • a. seripettor®: Cut the filling tube off according to the bottle height and attach it.
  • a’. seripettor® pro: Adjust the length of the telescopic filling tube to the bottle height and attach it.

    • a. Screw the instrument (GL 45 threads) onto the reagent bottle.
    • To avoid tipping over, use a bottle stand for small bottles.
    • NOTICE: For bottles with other thread sizes, select a suitable adapter, see Accessories/spare parts.
  2. Transporting the instrument
    • When installed on a reagent bottle, always carry the instrument as shown in the figure and always store it upright!
  3. Priming the instrument
    • a. Hold the discharge tube and remove the closure cap/ screw cap.
    • b. To avoid splashes, hold discharge tube orifice on the inner wall of a suitable receiving vessel. For priming, let the piston move up approx. 30 mm and push it down rapidly until the lower stop. Repeat this procedure approximately 5 times until there are no more air bubbles under the piston. Bubbles less than 1 mm are permitted.

NOTICE: Before using the instrument for the first time, ensure it is rinsed carefully and discard the first few samples dispensed. Avoid splattering.

Selecting the volume

NOTICE: Liquid is dispensed when setting the volume!

  • a. Hold the discharge tube and remove the closure cap/ screw cap.
  • b. Hold the discharge tube orifice on the inner wall of a suitable receiving vessel.
  • c. With one hand, press the actuating unit downward and hold it.
  • d. Place the receiving vessel under the discharge tube orifice.
  • e. Loosen the volume selector thumb screw by a half turn, slide the pointer vertically to the desired volume and retighten the volume thumb screw.

  • a. Hold the discharge tube and remove the closure cap/ screw cap.
  • b. Hold the discharge tube orifice on the inner wall of a suitable receiving vessel.
  • c. Push the piston down to the lower stop evenly and with minimal effort and then let it move up slowly.
  • d. Wipe off the discharge tube against the inner wall of the receiving vessel.
  • e. Seal the dispensing tube with the closure cap/screw cap.

Dispensing with the flexible discharge tube

The flexible discharge tube can optionally be used for serial dispensing (see Accessories/spare parts,). The specified accuracy and coefficient of variation of the instrument are only obtained for volumes > 2 ml and by gently approaching the upper and lower stops. The coil of the tubing can be stretched to a maximum length of 800 mm. The tubing should lie neatly in loops and must not be twisted. The parts which come into contact with media consist of borosilicate glass, Al2-O3-ceramic, ETFE, PTFE, platinum-iridium and PP.

Never use the flexible discharge tube for:

  • Liquids attacking borosilicate glass (e.g., hydrofluoric acid)
  • Peroxides, as these are decomposed catalytically by platinum-iridium (e.g., H2O2)

The operating exclusions for the respective instrument in use also apply.

Installing the flexible discharge tube

  • a. Screw the tube holder to the valve block and install the collection tube. seripettor®: Before installing the flexible discharge tube, remove the built-in elastomer sealing ring and replace it with the provided PTFE sealing ring.
  • b. Check that the flat seal is inserted in the adapter.
  • c. Slide the discharge tube with flexible tubing onto the adapter and tighten the cap nut by hand. Then slide the adapter onto the valve block and again tighten the cap nut by hand. Use a bottle holder.
Dispensing with a microfilter

Discharge tube with Luer-Lock attachment for microfilters

The discharge tube with Luer-Lock attachment for microfilters can optionally be used for sterile filtration of media, see Accessories/spare parts. The discharge tube with Luer-Lock attachment allows a microfilter to be connected for sterile filtration. The parts which come into contact with media consist of: borosilicate glass, l2O3-ceramic, ETFE, FEP, PFA, PTFE, platinum-iridium and PP (Luer-Lock attachment).

Therefore, never use for

  • Liquids attacking borosilicate glass (e.g., hydrofluoric acid)
  • Peroxides, as these are decomposed catalytically by platinum-iridium (e.g., H2O2)

The operating exclusions for the instrument and microfilter in use also apply. Only use microfilters with Luer-Lock. Never exceed the maximum operating force of 100 N.

  • Dismantle the standard discharge tube from the seripettor® pro and slide the discharge tube with Luer- Lock approx. 2 mm onto the adapter for discharge tubes.
  • Then firmly tighten the cap nut by hand. Commercially available sterile filters can be installed onto the Luer- Lock attachment. After connecting the filter, check that it is firmly seated.

NOTICE: Observe the regulations for handling sterile media. The increased flow resistance can lead to leaking liquid in the dispensing cartridge. To keep any liquid leaks to a minimum, we recommend dispensing with little force and using a filter with a large filter surface. Please follow the recommendations of the filter manufacturer in terms of media compatibility. Watch out for any increase in pressure during use, and change the filter in a timely manner before it becomes blocked.

Bottle stand

For small bottles, and when using the flexible discharge tube, use a bottle stand to prevent tipping over (Accessories/spare parts).

  • Position the mounting plate at the appropriate height.
  • As shown, insert the instrument firmly into the holder until it audibly clicks into place.
  • Then lock the holder into place with the screw.

Error limits

20 °C Ex: Error limits in relation to the nominal capacity (= maximum volume) indicated on the instrument at equal temperature (20 ºC/68 °F) of instrument, ambient environment and distilled water. Testing takes place according DIN EN ISO 8655- 6 with a completely filled instrument and with uniform and smooth dispensing.

NOTICE: The maximum error for a single measurement can be calculated from the sum of error limits EL = A + 2 × CV (e.g. for 10 ml: 120 μl + 2 x 20 μl = 160 μl).

Checking the Volume (Calibration)

Depending on use, we recommend that gravimetric testing of the instrument be carried out every 3-12 months. This time frame should be adjusted to correspond with individual requirements. The complete testing procedure (SOP) can be downloaded at www.brand.de. For GLP- and ISO- compliant evaluations and documentation, we recommend the EASYCAL™ calibration software from BRAND. Demo version can be downloaded from www.brand.de. Gravimetric volume testing according to DIN EN ISO 8655- 6 (for measurement conditions, see Error limits, p. 55) is performed as follows:

  1. Prepare the instrument
    • Clean the instrument (Cleaning, p. 57), fill it with distilled H2O and then prime it carefully.
  2. Check the volume
    • At 10 pipetting series with destilled H2O and weighings in 3 volume ranges (100 %, 50 %, 10 %) are recommended
    • For discharge, depress the piston slowly and steadily without force until the lower stop
    • Wipe off the tip of titration tube.
    • Weigh the pipetted amount with an analysis scale. (Please refer to the user manual of the scale manufacturer.)
    • Calculate the dispensed volume. The Z factor takes account of the temperature and air buoyancy.

Cleaning

WARNING: Components filled with reagent

  • The cylinder, valves, telescopic filling tube and discharge tube contain reagent!
  • Never remove the discharge tube while the dispensing cylinder is filled.
  • Point the valves and tube openings away from your body.
  • Wear protective clothing, eye protection and protective gloves!

The instrument must be cleaned in the following situations to assure correct operation

  • immediately when the piston is difficult to move
  • prior to a reagent change
  • prior to long term storage
  • prior to dismantling the instrument
  • prior to changing the valve
  • prior to autoclaving
  • regularly when using liquids which form deposits (e.g., crystallizing liquids)
  • regularly when liquids accumulate in the screw cap.
Basic cleaning

Completely emptying the instrument

  • Screw the instrument onto an empty bottle and empty it completely by dispensing. If the instrument is equipped with a recirculation valve, it must be emptied in both the ‘dispense’ and ‘recirculate’ settings.

Rinsing the instrument

  • Screw the instrument onto a bottle filled with a suitable cleaning agent (e.g. deionized water) and rinse the instrument several times by completely filling and emptying it.
Cleaning and replacing valves

Filling valve

  • a. Remove filling tube.
  • b. Unscrew the filling valve with a coin and extract the filling valve body.
  • c.  If the sealing ring is contaminated or damaged, carefully remove it using a pair of angled tweezers.
  • d. Clean any contaminated parts (e.g., in an ultrasonic bath).
  • e. Insert cleaned or new sealing ring.
  • f. Screw in the cleaned or new filling valve first by hand and then tighten it with a coin.

Discharge valve

  • a. Loosen the screw cap of the dispensing tube.
  • b. Extract the discharge valve.
  • c. If the sealing ring is contaminated or damaged, carefully remove it using a pair of angled tweezers.
  • d. Clean any contaminated parts (e.g., in an ultrasonic bath).
  • e. Install the dispensing tube with the cleaned or new discharge valve.

Filling valve

  • a. Remove filling tube.
  • b. Use the assembly tool to unscrew the filling valve.
  • c. If the sealing ring is contaminated or damaged, carefully remove it using a pair of angled tweezers.
  • d. Clean any contaminated parts (e.g., in an ultrasonic bath).
  • e. Insert cleaned or new sealing ring.
  • f. Screw in the cleaned or new filling valve first by hand and then tighten it with the assembly tool.

Discharge valve

The discharge valve is built into dispensing tube.

  • a. Dismantle the dispensing tube and clean in an ultrasonic bath if necessary.
  • b. Install cleaned or new dispensing tube.

Dislodging a stick valve ball

Replacing the dispensing cartridge

The dispensing cartridge is a wear part. In case of sluggishness or leakage between cylinder and piston, the dispensing cartridge must be replaced. To prevent injuries due to chemical exposure, cleaning must be performed before replacing the dispensing cartridge.

  • a. Screw the instrument onto an empty bottle and empty it completely by dispensing multiple times.
  • b. With one hand, press the actuating unit downward all the way and hold it. Loosen the volume selector thumb screw by a half turn, slide the pointer vertically to the upper stop and re-tighten the volume thumb screw.
  • c. Release the piston lock.
  • d. Unscrew and remove the actuating unit.
  • e. Unscrew the dispensing cartridge and replace it with a new one, see Accessories/spare parts.
  • f. Screw the actuating unit on and engage the piston lock.

Autoclaving

NOTICE

The following are not autoclavable: Dosing unit and actuator unit

The valve block, aspiration tube, dosing cannula and valves are autoclavable at 121 °C at 2 bar and aholding time of at least 15 minutes according to DIN EN 285.

  • a. Unscrew actuating unit and dispensing cartridge, see Replacing the dispensing cartridge.
  • b. Loosely seal the valve block with the closure cap (see Accessories/spare parts).
  • c. Insert the autoclavable filling tube (see Accessories/spare parts with the sealing ring up into filling valve.
  • d. Loosely screw the sealed valve block with filling tube and dispensing tube onto the filled bottle.
  • e. Seal the ventilation opening with an autoclavable membrane filter (0.2 μm).
  • f. Open dispensing tube, loosen cap nut.
  • g. Autoclave.

NOTICE

Allow parts to cool slowly after autoclaving Allow to cool in the closed autoclave to avoid deformation due to too fast cooling (cooling time approx. 2 hours, exception: seripettor® with agar culture medium up to max. 60 °C). After every autoclaving, inspect all parts for deformities or damage. If necessary, replace them. It is the user’s responsibility to ensure effective autoclaving.

Preparing for sterile work

  • a. After autoclaving, seal the dispensing tube and tighten the screws.
  • b. Under sterile conditions, remove the closure cap.
  • c. Screw in the sterile dispensing cartridge (see Accessories/ spare parts).
  • d. Fasten the actuating unit.

Troubleshooting

Dispensing not possible| Discharge valve stuck| Unscrew the discharge valve from the valve block, clean it and replace if necessary. If the valve ball is stuck, use a 200 µl plastic tip to dislodge it.
Air bubbles in the instrument| Reagent with high vapor pressure has been drawn in too quickly| Slowly draw in reagent.
Valve screw connections loose| Tighten the valves firmly with the assembly tool.
The instrument has not been primed| Prime the instrument, see First Steps
Filling tube is loose or dam- aged| Push the filling tube on firmly. If necessary cut off ap- prox. 1 cm of tube at the upper end and re-connect it or replace filling tube.
Valves not firmly connected or damaged| Perform cleaning, see Cleaning. Tighten the valves using the assembly tool.
Dispensed volume too low| Discharge tube is loose or damaged| Correctly install the discharge tube. Replace de- formed or damaged discharge tube.
Filling tube is loose or dam- aged| Perform cleaning, see Cleanin. Push the filling tube on firmly. If that is insufficient, cut off approx.

1 cm of tube at the upper end or replace the filling tube.

Filling valve is loose or dam- aged| Perform cleaning, see Cleaning. Tighten the filling valve. If necessary, replace the valve with seal- ing ring, see Cleaning and replacing valves,
Frequent dispensing > 40 ºC| Replace dispensing cartridge, see Replacing the dispensing cartridge.

  • When returning products, please enclose a general description of the type of malfunction and the media used. If information regarding media used is missing, the instrument cannot be repaired.
