In the early anagen phase, cells from the dermal papilla activate bulge stem
cells, inducing new hair follicles to grow and replace the old hair that
sheds. The bulge stern cells then self replicate and migrate into the matrix

The derma papilla fibroblasts and epithelial stem cells need to communicate
together in order to trigger the hair follicle’s cycle of growth. Otherwise,

Abstract
Adenosine is an effective treatment for androgenetic alopecia (AGA) in
Japanese men and women. Adenosine exerts its effects by significantly
increasing the proportion of thick hair. In this study, we assessed the
clinical outcome of adenosine treatment for 6 months in 38 Caucasian men. The
change in the proportion of thick hair (60 pm) compared with baseline in the
adenosine group was significantly higher than that in the placebo group (P c
0.0001). The change in vellus hair proportion (<40 pm) was significantly lower
in the adenosine group than that in the placebo group (P = 0.0154). The change
in hair density compared with a baseline of the adenosine group was also
significantly higher compared with that of the placebo group (P = 0.0470). No
adverse effects due to treatment were noted during this study by
dermatological evaluation. Adenosine is effective In increasing the proportion
of thick hair in Caucasian men with AGA as well as in Japanese men and women.

Abstract
Objective: Hair thickness is more important than hair density in the
appearance of baldness in male with androgenetic alopecia (AGA). Adenosine
improves hair loss by stimulating hair growth and by thickening hair shafts in
women. The objective of this study was to evaluate the hair growth efficacy
and safety of topical adenosine in men with AGA.
Methods: A lotion containing either adenosine or niacinamide was
administered to the scalps of 102 Japanese men twice daily for 6 months in a
double-blind, randomized study. Efficacy was evaluated by dermatologists who
assessed the quality of the hair and by calculating the percentages of vellus-
like and thick hairs among the vertex hairs, as well as hair density.
Results : Adenosine was significantly (P < 0.05) superior to niacinamide
in terms of global improvement of AGA, increase in the percentage of thick
hairs (at least 60 pm) and self-assessment of hair thickness by the study
participants. No causal adverse event due to the adenosine lotion was
observed.
Conclusion: These data indicate that adenosine increases thick hair ratio
in Japanese men with AGA, and this compound is useful for the improvement of
AGA.

Abstract
Hair regress : on and balding are distressing concerns for an increasing
number of people due to changes in lifestyle and serious nutritional
imbalances. Therapies for the treatment of hair loss are needed. Among
potential therapeutics, adenosine has been suggested as a potent regulator of
hair growth. In this study, we investigated the effects of adenosine on hair
follicles and dermal papilla (DP) cells, and the mechanism underlying the
action of adenosine. Hair follicles are organs, including DP cells that are
responsible for the production of hair fibers by inducing and maintaining the
hair growth phase (anagen). In a culture of DP cells in vitro adenosine
stimulated the proliferation of DP cells by increasing thymidine uptake.
Subsequently, adenosine activated and elongated the anagen phase by increasing
the uptake of radiolabeled cysteine in an organ culture of mouse vibrissae
hair follicles. We also confirmed that adenosine promoted the expression of
several growth factors that are responsible for hair growth, including
fibroblast growth factors (FGF)-7, FGF-2, insulin-like growth factor (IGF)-1,
and vascular endothelial growth factor (VEGF) in a cDNA microarray with semi-
quantitative RT-PCR. Transcriptional activation of 13-catenin in DP cells was
increased by adenosine in a luciferase assay. p-catenin is a co-activator of
Wnt/P-catenin signaling that induces morphogenesis and differentiation of hair
follicles and also acts to transactivate downstream signaling pathways,
including the ERK pathway. Using Western blotting, we found that adenosine
stimulated phosphorylation of ERK. CREB and AC. These results suggest that
adenosine stimulates the growth of hair follicles by triggering the expression
of growth factors and 13-catenin, and by inducing their downstream target
signaling pathways.

Strengthening hair with Centella Asiatica: a report of clinical and
subjective efficacy of local treatment with a 0.5% hair lotion

BACKGROUND : Hair follicles represent a regenerating organ, surrounded by
a dense capillary network and sebaceous gland, with some capillaries
penetrating to perfuse also the dermal papilla that promotes follicular
growth. Centella Asiatica is an Ayurvedic herb of the Umbellifere family,
containing high levels of the glycosylated triterpenes
Madecassoside and Asiaticoside, are known respectively to facilitate collagen
III and boost collagen I synthesis at the perivascular tissue level. These
properties improve follicle perfusion and metabolism, and thus extracts from
Centella Asiatica may be helpful to strengthen and invigorate hair, with a
strong potential for hair care applications. METHODS: Our study evaluated
these targets by comparing the efficacy and tolerance/appreciation of a hair
lotion containing Centella Asiatica extract (Centerox®), versus a placebo
lotion, in 30 volunteers with telogen effluvium or mild to moderate alopecia,
15 per group. The volunteers used 2 ml of the assigned lotion each day over a
period of 4-8 weeks. Efficacy was evaluated by means of the pull test and wash
test, whereas tolerance was self-assessed with a questionnaire. Additionally,
an in vitro study was performed to evaluate the ability of Centella Asiatica
triterpenes to improve epidermal cohesion and dermal-epidermal junction on
epidermal keratinocytes. RESULTS: The lotion containing Centella Asiatica
allowed a 41% decrease in hair loss in the washing test, and a small increase
in hair strength in the pull test (P<0.05). The product was well tolerated. A
33% increase in laminin-5 expression was also observed in the in-vitro study
(P<0.05). CONCLUSIONS: Centerox® is effective and well-tolerated in the
treatment of alopecia, resulting in improved hair strengthening and decreased
hair loss. Moreover, triterpenes upregulate laminin-S, responsible for hair
growth, due to their action in cell differentiation, migration, adhesion, and
phenotypes expressions.

Ubiquinones are the most widespread and therefore best-investigated quinones.
Due to their hydrophobic isoprenoid side chain, ubiquinones can be solubilized
in organic solvents or lipids but are insoluble in water. Using a specific
emulsifier system it has been possible to deliver positive effects to
biological systems also from aqueous formulations. Ubiquinone-50, also
referred to as coenzyme Q10, is well known in cosmetic science and especially
in skincare because of its antioxidant activity. It is found in the membranes
of peroxisomes, lysosomes, vesicles, the endoplasmic reticulum, and notably in
the inner membrane of the mitochondrion, where it is an important part of the
electron transport chain. Using Coenzyme Q10 in the proposed emulsifier system
we could show that the molecule not only has relevance as an anti-aging
bioactive in skincare but also has positive effects on the human hair
follicle. The hair follicle is a complex mini-organ and the synthesis of hair
keratin, the major component of hair fibers, is an essential prerequisite for
the growth of strong and healthy hair. But like all biological systems the
hair follicle, the biologically active part of the hair, also undergoes an
aging process associated among other things with a decline in certain hair
keratins. Due to this age-related shift in basic structural proteins of the
hair shaft, mature hair often becomes fragile and difficult to manage.
Therefore it is a challenge for cosmetic science to provide bioactive to fight
age-related changes and maintain a youthful appearance of hair. Using
cultivated hair follicle keratinocytes we identified coenzyme Q10 as a potent
bioactive that stimulates the gene expression of different hair keratins,
especially those which are reduced during aging processes in hair follicles.
These results led us to investigate a shampoo and a tonic formulation enriched
with coenzyme Q10 in a placebo-controlled panel study. In a left/right
comparison, a group of healthy volunteers older than 40 years of age applied
the formulations daily for 4 days. Throughout the test period, the gene
expression of different hair keratins from plucked hair follicles was
determined using quantitative polymerase chain reaction techniques. Subsequent
statistical analysis revealed an increase in age-relevant hair keratins in
human hair roots treated with coenzyme Q10, thus pointing out the striking
benefits of coenzyme Q10 in hair care formulations. We conclude that coenzyme
Q10 is an ideal ingredient for hair care formulations, providing anti-aging
properties through the activation of specific keratins aligned with the needs
of mature hair.

Abstract
The scalp is exposed to environmental hazards including airborne pollutants which exert adverse effects on skin health Therefore compounds for defending skin from pollutants have attracted interest in the cosmeceutical community We investigated whether Ecklonia cave exhibited prophylactic effects against urban pollutants by measuring cell viability and cell cycle distribution in human follicle dermal papilla cells (HFDPC) The effect of E. cave on pollutant-induced damage to skin barrier was determined by measuring filaggrin and MMP-1 expression in both keratinocytes and in a skin explant model In a clinical trial, the effect of E cave on scalp skin of patients with scalp scale was observed by evaluating hydration and redness after 4 weeks of daily treatment with a shampoo containing E cave extract E cave extract recovered the loss of cell viability and abnormal cell cycle distribution induced by urban pollutants in HFDPCs It also attenuated pollutant-induced damage to skin barrier by decreasing MMP-1 and increasing filaggrin expression in keratinocytes and the epidermis of skin explants Moreover E cave showed soothing effects on human scalp by increasing hydration and decreasing redness in a clinical trial Collectively. This study was conducted to evaluate the effect of Ecklonia cava. a marine
alga native to Jeju Island in Korea. on the promotion of hair growth. When
vibrissa follicles were cultured in the presence of E. cava enzymatic extract
(which contains more than 35% of the director) for 21 days. E. cava enzymatic
extract increased hair-fiber length. In addition, after topical application of
the 0.5% E. cava enzymatic extract onto the back of C57BL ‘6 mice. anagen
progression of the hair shaft was induced. The treatment with E. cava
enzymatic extract resulted in the proliferation of immortalized vibrissa
dermal papilla cells (DPC). Especially. director. among the isolated compounds
from the E. cava enzymatic extract. showed activity that increased the
proliferation of DPC. When NIH3T3 fibroblasts were treated with the E. cava
enzymatic extract and the isolated compounds from the E. cava enzymatic
extract. the E. cava enzymatic extract increased the proliferation of NIH3T3
fibroblasts. but the isolated compounds such as nickel. director.
phloroglucinol and triphlorethol-A did not affect the proliferation of NIH3T3
fibroblasts. On the other hand. the E. cava enzymatic extract and dieckol
significantly inhibited 5a-reductase activity. These results suggest that
directors from E. cava can stimulate hair growth by the proliferation of DPC
and/or the inhibition of 5a-reductase activity.
