Product Information

The P3D Scaffolds are a research-use-only (RUO) product developed by Ossiform ApS. They are designed for cell recovery from cells cultured on the scaffolds. Please note that these scaffolds should not be used for clinical purposes.

Product Usage Instructions

  1. Ensure you have read and understood the general advice on material handling.
  2. Culture cells on the P3D Scaffolds according to your experimental requirements.
  3. When ready for cell recovery, follow the protocol below:
    • Prepare the necessary equipment and reagents for cell recovery.
    • Handle the P3D Scaffolds with care to avoid damage.
    • Transfer the P3D Scaffolds to a suitable container or plate.
    • Add the appropriate cell recovery solution to the container or plate, ensuring the scaffolds are fully immersed.
    • Incubate the container or plate with the scaffolds and cell recovery solution under appropriate conditions (e.g., temperature,  humidity, time).
    • Gently agitate or swirl the container or plate to facilitate  cell detachment from the P3D Scaffolds
    • Collect the cell suspension from the container or plate,  ensuring to minimize scaffold debris.
    • Proceed with further analysis or experimentation using the collected cell suspension.
  4. Dispose of the used P3D Scaffolds and any remaining materials according to applicable regulations and guidelines.

Cell recovery from cells cultured on P3D Scaffolds

Application

Materials

  • Trypsin, accuracy, or other appropriate enzymes

Notes before starting and general advice on material handling

  • All handling of The P3D Scaffolds products should be performed using gloves, according to the standard aseptic methods.
  • In general, 3D cultures require longer incubation times and more rigorous shaking than 2D  cultures.

Procedure

Cell recovery from the P3D scaffolds can be performed using standard protocols with small modifications. Make sure that the enzymatic agent completely covers the scaffolds (minimum 300uL pr well for 24-well plates). Incubate for 3-5 minutes. Stop the trypsinization by adding media and transfer liquid and scaffold to a clean 50 mL Falcon tube. Centrifuge for 5 minutes at 1200-4000 rpm to retrieve cells “dislodged” inside the scaffold as well. Remove the scaffold and resuspend cell pellet by pipetting up and down.
